Eukaryote diversity of typical grassland soil based on 454 pyrosequencing

  • Eukaryote diversity of typical grassland soils with different enclosure stages was analyzed using 454 pyrosequencing. The results showed that soil eukaryotes, including microorganisms, animals and plants could be amplified and found using selected primers in V4 regions. The soil mycoflora is mainly composed of Ascomycota, Chytridiomycota, Basidiomycota, Chlorophyta, Glomermycota and Protecobacteria; Animal community is mainly composed of Craniata, Arthropoda, Nematoda and Annelida; Plant community is mainly composed of Embryophyta and Liliopsida. Under different taxonomic levels, there are different dominant floras for mycoflora, but only differences among species for animal community, in grasslands with different enclosure stages. Rational usage of enclosure grasslands could be conducted based on combining soil microorganisms with restoration years of soil. 454 pyrosequencing can be used on researches of unknown soil animals, plants and microorganisms.
